Pristimerin Contributes to Gefitinib Resistance in Lung Cancer Cells by regulating microRNA-936 expression

To investigate the effect of pristimerin on gefitinib resistance in lung cancer cells and its regulation microRNA-936. Lung cell HCC827 was cultured vitro, resistant HCC827/gefitinib established were randomly assigned to control group, pristimerin-L pristimerin-M pristimerin-H gefitinib+pristimerin gefitinib+microRNA-negative gefitinib+microRNA-936 gefitinib+pristimerin+anti-microRNA negative group gefitinib+pristimerin+anti-microRNA-936 group. 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide used detect inhibition rate proliferation, as well median half-maximal inhibitory concentration; expression amount microRNA-936 detected by quantitative reverse transcription-polymerase chain reaction; migration invasion transwell chamber assay. Compared with cells, proliferation significantly lower concentration value higher (p<0.05); compared increased, decreased increased (p<0.05) group; number gefitinib+microRNA showed numbers (p<0.05). Pristimerin may enhance sensitivity inhibiting up regulating expression.
